硅橡胶控释的硝酸甘油经皮缓释剂的研制
PreParation and in vitro Release of Silicone Nitroglycerine Transdermal TheraPeutic System
【摘要】 用硅橡胶与硝酸甘油、助剂混合,经真空排泡,加温固化则得基质型硝酸甘油缓释剂。其内部结构为非均相体系,微粒粒径在20~200微米之间。用Franz细胞扩散仪,以生理盐水为缓释接受液,在37±0.5℃恒温与电磁搅拌下,进行体外释放规律的测定。结果证明:6小时后缓释作用接近零级释放;通过回归分析得6—24小时间的直线方程为(?)=132.7(微克/厘米~2·小时)。
【Abstract】 Silicone, nitroglycerine, and additives were mixed, exhausted under vacuum and vulcanized at 60℃ temperature. A heterogeneous siliconenitroglycerine matrix system was obtained. The diameters of the microsealed compartments in the system were from 20 to 200μm. The system releasing nitroglycerine into 0.9% NaCl/H2O (W/W). at 37-0.5℃ under stirring was studied with Franz cell diffusion device. The relationship of the releasing rate to the releasing time showed that releasing rate approximately corresponded to zero order releasing after 6 hrs. Least squares analysis was used to locate the best lines through these. The regression equation obtained is 132.7(μg/cm2·hr).
