IL2吸收试验检测CTLL2细胞和Con A活化-小鼠脾细胞的IL2受体
ABSORPTION TEST FOR THE DETECTION OF IL2 RECEPTORS ON IL2 DEPENDENT CELL LINE CTLL2 CELLS AND CON A ACTIVATED-MOUSE SPLEEN CELLS
【摘要】 用初步纯化的白细胞介素2(IL2)建立了检测IL2受体的实验方法——IL2吸收试验,以IL2吸收率表示IL2受体的数量。IL2依赖细胞株(CTLL2)细胞和Con A活化一小鼠脾细胞有能力吸收除去,而正常休止状态小鼠脾细胞和未经con A刺激的脾细胞不能吸收除去上清液中的IL2活性。此外,IL2的吸收与CTLL2细胞的数量有关,细胞数量为8×10~6/ml时几乎吸收除去上清液中全部IL2活性,随着细胞数量的减少,IL2吸收率随之降低。结果证实IL2吸收试验检测IL2受体有特异性强,稳定,半定量和可重复等特点。
【Abstract】 We developed an interleukin 2 (IL2) absorption test to detect IL2 receptors on the surface of lymphocytes. The IL2 dependent cell line OTLL2 cells and Con A-activated (but not resting) mouse spleen cells could absorb IL2 activity from the supernatants. In addition, IL2 removal by OTLL2 cells was dependent on cell numbers. 8 × 106/ml cells completely absorbed IL2 activity out of the supernatants, absorption decreased with decreasing numbers of OTLL 2 cells. The results indicate that the IL2 absorption test is specific for IL2 receptors and is quantitative and reproducible.
