NaCl及γ射线对小麦愈伤组织某些生理指标的影响

【摘要】 利用鲁麦10号小麦品种幼穗作为外植体诱导愈伤组织。研究了NaCl及γ射线对小麦愈伤组织的生长、脯氨酸含量及细胞形态大小的影响。结果如下:1.γ射线及NaCl都明显的抑制愈伤组织的生长。0.3%的NaCl和5KR的照射量是愈伤组织生长的临界值;30KR是致死的起始照射量:2.NaCl对愈伤组织的脯氨酸含量也有显著的影响,二者呈抛物线相关。γ射线使脯氨酸含量升高,但超过5KR,脯氨酸含量又呈下降的趋势:3.γ射线处理愈伤组织能使细胞的长及宽都增大,并产生巨型细胞,加速愈伤组织的老化。NaCl对愈伤组织细胞大小无显著影响,但能引起细胞大小变异系数的增加。

【Abstract】 Wheat immature inflorescence was used as explant for in vitro culture. The effects ofy-rays and NaCl on the callus growth, content of proline and cell shape and size were in-vestigated. The results were as follows: 1. Growth and developmet of the callus was inhibited by r-rays and NaCl. The expo-sure dose of 5KR and 0.3% NaCl concentration could be considered as the critical valueof callus growth and development. 30KR was the initial lethal dose of wheat callus. 2. The proline content of callus raised under salt stress, but it fell when the NaCl con-centration increased to some value. The relation of proline content and NaCl concentrtionshowed a parabolic curve. 0-5KR of r-rays caused the increase of the proline content, butproline content fell if the exposure was over 5KR. 3. y-rays not only caused the increase of the cell size and resulted in producing hugecells, but also, sped up the senility of the callus. NaCl did not affect the size of cells, but in-creased the variation coefficient of cell size.
