平颏海蛇蛇毒胰蛋白酶抑制剂

【摘要】 从平颏海蛇(Lapemis hardwickii)蛇毒中分离并纯化得到胰蛋白酶抑制剂。用咬皿法从活蛇收集到的蛇毒,经Sephadex G-75柱(2.1X93cm)凝胶过滤。得到5个组分;S-IS-Ⅱ,S-Ⅲ,S-Ⅳ,S-Ⅴ。其中S-Ⅴ组分为胰蛋白酶抑制剂,比活力为784单位/mg蛋白,是粗毒的16.9倍。在pH4.3,用浓度为15%(重量/体积)的聚丙烯酰胺的连续凝胶进行电泳时,它呈现单一的带。S-Ⅴ组分冻干后重27mg,占蛇毒总量的1.94%。

【Abstract】 The trypsin inhibitor was purified from the venom of Lapemis hard-wickii. By Sephadex G-75 column chromatography the fractions S- Ⅰ, S-Ⅱ, S-Ⅲ, S-Ⅳ, S-Ⅴ were obtained, among which fraction S-Ⅴ was a trypsin inhibitor containing 784 BAEE units/mg,which was 16.9 times the crude venom. By pH 4.3 and 15% polyacrylamide electrophoresis, it appeared as a single band. S-Ⅴ was 27mg, accounting for l.94% of the total of crude venom.
