人血管内皮细胞的体外培养及其特性
Culture and Properties of Human Endothelial Cells in Vitro
【摘要】 用胶原酶消化法定内皮细胞从人脐带静脉内膜分离,并培养在含有内皮细胞生长因子的RPMI1640培养基内,细胞在体外培养中可以传至第6—10代。细胞呈相互密切接触地单层生长。细胞大,呈多边形,且在培养中可以形成毛细血管芽。用单克隆抗体方法检查,胞浆第因子呈阳性反应。在透射电镜下于胞浆内查见WeibeI—PaIade氏小体。内皮细胞的培养有利于研究癌转移及血管疾病。
【Abstract】 Endothelial cells which were isolated from interior of human umbilical vein by collagenase digestion and cultured in medium RPMI 1640 with growth factor of cndothelial cells could be propagated to 6-10th pasage in vitro. The cells grew as monolayer and were closely contacted one an other. They were large and polygonal in shape and could from capillary buds in culture. Factor Ⅷ was positive in the cytoplasm with monoclone antibody method. The Weibel-palade’s bodies could be seen in the cytoplasm by TEM. The culture of endothelial cells will be useful in the research of cancer metastasis and vascular diseases
