【Abstract】 Birbeck granule is an adsorptive endocytic organelle which might be involved in antigen presentation. It was originally discovered in the epidermic Langerhans cells. Later, granules of the same type have been found in the veiled cells in afferent lymph and in the interdigitating cells of thymus and lymph nodes draining skin. But so far the granules have not been demonstrated in the dendritic cells of the spleen. By using the method designed by Kamperdijk et al, with some modifications, we isolated dendritic cells from rat spleen and the fractions containing enriched dendritic cells were obtained. Under transmission electron microscope, we found that there were cells containing Birbeck granules in the fraction. The morphology and structure of the cells were similar to those of dendritic cells described by Steinman. The granules were rod in shape with a limiting membrane and an electrondense inner core, scattered in the cytoplasm. From this study, it was confirmed that there were cells containing Birbeck granules in the rat spleen. This might be of significance in explaining the presence of immune response in the rat spleen.更多还原
