大鼠脾内含Birbeck颗粒的细胞
CELLS CONTAINING BIRBECK GRANULES IN RAT SPLEEN
【摘要】 Birbeck颗粒是一种吸附、内包的细胞器,可能与抗原的提呈有关。它们最初被发现于表皮的郎格罕细胞中,以后又见于输入淋巴的面纱细胞以及胸腺和引流皮肤淋巴结的交错网状细胞。但至今尚未确认脾内有含Birbeck颗粒的细胞。我们采用Kamperdijk分离大鼠树状突细胞的方法(稍加改良)分离大鼠脾细胞,获得了富有树状突细胞的组分。电镜下,这些细胞的形态和结构与Steinman等描述的树状突细胞相似。少数细胞内含有Birbeck颗粒。颗粒呈棒状,外有一层界膜,内有一条电子致密的中柱,散布于细胞质内。本研究证明大鼠脾内有含Birbeck颗粒的细胞。
【Abstract】 Birbeck granule is an adsorptive endocytic organelle which might be involved in antigen presentation. It was originally discovered in the epidermic Langerhans cells. Later, granules of the same type have been found in the veiled cells in afferent lymph and in the interdigitating cells of thymus and lymph nodes draining skin. But so far the granules have not been demonstrated in the dendritic cells of the spleen. By using the method designed by Kamperdijk et al, with some modifications, we isolated dendritic cells from rat spleen and the fractions containing enriched dendritic cells were obtained. Under transmission electron microscope, we found that there were cells containing Birbeck granules in the fraction. The morphology and structure of the cells were similar to those of dendritic cells described by Steinman. The granules were rod in shape with a limiting membrane and an electrondense inner core, scattered in the cytoplasm. From this study, it was confirmed that there were cells containing Birbeck granules in the rat spleen. This might be of significance in explaining the presence of immune response in the rat spleen.
