【摘要】 作者在细胞培养中发现田基黄对Ⅱ型单纯疱疹病毒(HSV-2)的复制有明显的抑制作用。它对 Vero-E6细胞的无毒浓度为7.5mg/ml(即相当于每毫升7.5mg 生药),最小有效抑制 HSV-2复制的浓度为2.5mg/ml。抑制指数[即病毒时照的滴度(对数)减去药物处理过的病毒滴度(对数)]为3.25,抑制明显。有效浓度的田基黄(2.5mg/ml)在细胞外对 HSV-2无直接灭活作用。HSV-2在复制周期中的早期即病毒的吸附或穿入阶段对田基黄敏感。此药在2.5mg/ml 的浓度下,抗 HSV-2的作用不亚于环胞苷(5ug/ml)或疱疹净(100ug/ml).因此,提出了用此药预防宫颈癌或与无环乌苷合用治疗HSV-2感染的可能性。更多还原

【Abstract】 Hypericum japonicum was found to be highly inhibitory to the replication of HSV₂ in tissue culture system.The non-cytotoxic concentration for vero-E6 cells is 7.5mg/ml（this concentration ia epuiva- lent to 7.5 mg of the original herbs）.The minimal complete inhibitory concentration for HSV₂ replication is 2.5mg/ml.The inhibitory index（i.e.the titer of virus control expressed in log TCID50 minus the virus titer of the drug treated sample expressed in log TCID50）is 3.25,indicating that the inhibition is very prominant.No direct inactivation of HSV₂ was observed when virus suspension was mixed with the effective inhibitory concentration of the drug（2.5mg/ml）.The critical time for complete inhibition of HSV₂ replication by Hypericum japonicum was at the beginning of virus adsorption（i.e.the“O”minute in the experiment）.The addition of the drug 15,30 minute and 1-12 hours after adsorption resulted only in partial inhibition indica- ting that Hypericum iaponicum exerted its maximal inhibtiory effect only during the stage of virus adsorption-penetration. In comparison with cyclocytidine or iodoxuridine,Hypericum japonicum exerted a greater anti-HSV₂ effect.The possible use of this drug in the prevention of cervical carcinoma and its use in combination with acyclovir for the treatment of HSV₂ infection is suggested.更多还原
