用氨气敏电极测定羊水腺苷脱氨酶活性

【摘要】 羊水腺苷脱氨酶(ADA)活性的测定有助于产前检查重症联合免疫缺陷症,但羊水中ADA活性偏低,难于直接检出。作者报道以氨气敏电极,应用标准加入法测定羊水中ADA活性。此法简便、快速,8 个不同样本测定的回收率为100.89±1.80%;7次测定的变异系数为1.64%。对62例13~41周正常妊娠羊水测定,ADA活性均值为0.267±0.173u(M±SD)。

【Abstract】 Adenosine deaminase (ADA) deficiencyis associated with an autosomal recession ofsevere combined immunodeficiency diseaseand is inherited. Setting up a method forthe prenatal determination of ADA activityin amniotic fluid is of great significance foreugenetics. This paper describes a method for deter-mination of the activity of ADA in amnioticfluid by an ammonia gas-sensing electrode.The average recovery (8 times) and C.V. (7times) are 100. 89±1. 80% and 1. 64% respec-tively. ADA activity in the amniotic fluidmeasurement was carried out in 62 nor-mally pregnant women at 13th-41st week,an average value and standard deviation is0.267± 0. 173 units. There is no relationshipbetween ADA activity and the pregnantperiod.
