~3H-环丁砜经口染毒后在大鼠体内的毒代动力学
STUDIES ON TOXICOKINETICS OF ~3H-SULFOLANE IN RAT AFTER ORAL ADMINISTRATION
【摘要】 作者采用放射性同位素技术,以~3H-环丁砜经口一次染毒后,对环丁砜在大鼠体内的吸收、血液及组织中的浓度、孕鼠及胎鼠体内组织分布,以及经胆汁、尿、粪排泄过程进行了研究。结果表明,环丁砜灌胃染毒后,吸收较快,并可广泛地分布于全身,主要随尿排出,且可通过血脑及胎盘屏障。
【Abstract】 In this paper, the absorption, distributionand excretion of 3H-sulfolane in rats werestudied by radioactivity trace method. The results showed that the curve ofblood 3H-sulfolane concentration versus timefitted a one-compartment open model withfirst order rate by curve fitting, statisticsand computer. The toxicokinetic parametersof 3H-sulfolane in blood were as follows:ka4. 48h-1. ke 0. 025h-1, T1/2ka 0. 15 h, T1/2ka27.37 h, AUC 1667.89 ng/ml·h, CL 24.47ml/h, Vd 4.84 L/kg, Tmax 1. 16h andCmax 41.01 ng/ml. The experiment showed that sulfolane was absorbed rapidly and completely by thegastrointestinal tract. The small intestinewas the major part of absorption.The levelof radioactivity was the highest in the liver,then the levels of radioactivity in the kidney,lung, thyroid gland, adrenal gland, pancreas,spleen, heart, muscle, brain, testis and fatdecreased gradually. 3H-sulfolane easilypassed through the blood-brain barrier andplacenta barrier. 3H-sulfolane was eliminated in urine andfaeces, especially in the former, ke of urine,and feaces were 0. 01899 and 0. 01172 respec-tively. The half-life periods of urinary andfecal elimination were 36.5h and 59h.
