不同大小单层脂质体的制备及其对细菌HCG结合物质(受体)重组的影响

【摘要】 脂质体(Liposome)已广泛应用于生物膜结构与功能的研究。作者采用超声法分别以超声时间10分钟、30分钟及2小时制备了脂质体。经电镜证实并测定了脂质体大小。通过凝胶过滤(Sepharose4B及Sephacry1 S-200)可见,随超声时间延长,其B峰增大而A峰消失。细菌HCG结合物质(受体)重组结果表明:不同大小的脂质体均有蛋白质嵌入颗粒,蛋白/磷脂比值超声10分钟者小于超声2小时者,而结合活性则前者明显大于后者。

【Abstract】 Liposome is widely used to study thestructure and function of cell membrane.Liposomes were prepared by using sonicatedphospholipid for ten mins, thirty mins andtwo hours. The vesicles were verified andthe size estimated by electron microscopy.Gel chromatograph of Sepharose 4B andSe hacryl S-200 allowed the separation ofa range of unilaminar particles from a soni-cated suspension. In column fractionationspeak B increased and peak A decreased intwo hours of sonicated time. The resultsreconstitution of HCG binding substancesfrom psendomonas maltophilia showed theinverted micelles (lipidic particles) observedby electron microscopy. In comparison ofthe sonification for ten mins with that fortwo hours,the ratio of protein to phospho-lipid in the former was lower, but itsbinding activity was higher than that of thelatter.
