急性髓细胞白血病患者血浆中硫鸟嘌呤的测定及药代动力学研究
DETERMINATION OF PLASMA CONCENTRATION AND PHARMACOTINETICS OF 6-THIOGUANINE IN ACUTE MYELOID LEUKEMIA
【摘要】 采用荧光法测定了18例用硫鸟嘌呤化疗的急性髓细胞白血病(急髓白血病)病人的血药浓度,按口服一室开放模型拟合后进行非线性迭代回归并计算药代动力学参数。t1/2a:95min;t1/2:530min;Tmax(达峰时间):272min;Cmax(峰浓度):40ng/ml。
【Abstract】 A sensitive fluorometric method is described for determination of 6-thioguanine (6TG) in plasma. It is based on that thioguanine can be oxidized to sulphonate product with high fluorescence. The exitcation wavelength is 330nm and the emission wavelength is 410nm. The lowest limit of detection is 5ng/ml (plasma) and recovery was about 85%. Plasma levels of 6.TG in 18 patients with acute myeloid leukemia(AML) had been measured after a sigle oral dose (2.53.0mg/kg/d) of 6TG. The concentration-time course fits one compartment open model. The concentration -time curve was calculated with non-linear iteration regression by computer. The pharmacokinetic parameters showed that t1/2a, t1/2a Tmax are 195 min, 530 min, and 272 min respectively, and Cmax is about 40ng/ ml.
