血小板过氧化物酶的电镜观察诊断巨核细胞性白血病

【摘要】 在光镜下难以将巨核细胞性白血病的原始细胞与其它急性白血病的原始细胞,特别是小原淋细胞相区别。血小板过氧化物酶(PPO)出现在巨核细胞系早期,而且有特异性。此酶只有在电镜下才能观察到。本文用预固定法显示PPO,阳性率高,简便可靠,并初步诊断了一例巨核细胞性白血病。PPO方法可以早期诊断巨核细胞性白血病,对鉴别其它急性白血病有重要意义。

【Abstract】 It is difficult to distinguish megakaryoblasts from small lymphocytes and undifferentiated celis with light microscope.Platelet Perosidase (PPO), which is recognized to be the particular enzyme mark in the megakaryocyte system, can be used specifically to diagnose megakaryoblastic leukemia with electron microscope.The method of prefixation, one of the three methods reported in foreign litefatures, proved to be higher in. sensitivity, better in keeping morphology, simpler in process, with which we have succeeded in diagnosing a case of megakaryoblastic leukemia.The following is an introduction to the method.The celis were prefixed for 30 min in 0.1% glutaraldehyde and then incubated in the dark for 60 min in 20mg DAB/0.1mol/L Tris-HCl-NaCl 10ml containing 0.003% H2O2. After washing in Ringer-Tris-HCl, the celis were fixed in 1.25% glutaraldehyde and ppstfixed in 1% osmium tetroxide.Our results show that dense PPO reaction products could be observed in some particular places of the megakaryoblasts such as nuclear envelope and endoplasmic reticulum but not in Golgi apparatus and granules.
