【Abstract】 Rhodamine 6GDN, a fluorescent dye, was found to inhibit cultured human gastriccarcinoma cell (SGC-7901) growth in vitro by 50%at 6.5ng/ml within 24hr of exposure.The use of 1μg/ml Rhodamine 6GDN, a non-growth inhibitory concentration, incombination with lower power Argon laser radiation produced a remarkable cytotoxicityon SGO-7901 cel1s. After 1-4 hr of radiation, the mitotic cells were decreased significantlyand the morphologic changes such as disarrangement and aberrant mitosis of cells,pyknosis, coagulation and fragmentation of the nuclei were observed. Twenty-four hourslater, 98% of cells treated with Rhodamine 6GDN-Ar~+ laser were killcd. A study of~8H-TdR incorporation into DNA of cells showed that the% inhibition of the precursorincorporation was 66% when the cells were pretreated for 24 hr with Rhodamine 6GDN andthen radiated with Ar~+ laser, whereas treatment with Rhodamine 6GDN alone decreasedthe 8~H-TdR incorporation by less than 40%. Using colony formation method,the viabilityof Rhodamine 6GDN on the normal hamster lung cells(V79) and malignant cells SGO-7901 was measured. The results showed that the compound possessed a selective cytotoxiceffect on malignant cells.
