人乳头瘤病毒和人子宫颈鳞状上皮细胞癌关系的超微结构与基因分子
Gene Molecular and Ultrastructural Studies on Relationship between Human Squamous Epithelial Carcinoma of Uterine Cervix and Human Papilloma Virus
【摘要】 以人乳头瘤病毒(HPV)DNA为探针,通过核酸杂交方法,检测宫颈癌病人活体细胞DNA中的病毒相关序列,用透射电镜观察癌细胞的形态特点。32例宫颈癌组织中23例与HPV16型探针杂交呈阳性反应,阳性率为72%。杂交阳性的癌细胞核内小体的出现率增加。
【Abstract】 Thirty- two biopsies from patients with squamous epithelial carcinoma of uterine cervix were collected from Beijing districts. All samples were separated into two parts. One part was used for DNA hybridization, HPV-6B , 11 , 16 and 18 plasmids DNA being used as probes and labeled by 32P-dTTP with nicktransla-tion method. DNA extracted from biopsies were spotted on nitrocellulose paper. 2 μg per dot, 2 μg DNA of salmon sperm being used as negative control and 1 ng of each HPV-plasmid as positive control. It was found that 23 out of 32 cervical carcinomas contained HPV-16 specific sequences, 3 out of 32 revealed HPV-18 DNA gene sequence and there were no positive hybridization with HPV-6B and 11 in all biopsies. Another part was used for ultra-structural research. The results reveaeld that the nuclei of cancer cells appeared swollen with heterochromatin less than normal cells. Two kinds of granules appeared in nuclei, one was interchromatin-granules 300 A in diameter , occasionally over ten of them aggregated into clusters and distributed among euchromatin. Another was perichromatin granules 500-800 A in diameter, always dispersed singlely in heterochromatin. Furthermore, a different type of nuclear body 0.01-0.05 μm in diameter also appeared in nuclear matrix. They were round or oval in shape and consisted of fine fibri-lles arranged in a concentric circle, sometimes there were several dense granules at the center of the concentric circle. These ultrastructural characteristics were more significent in those samples with positive hybridization of HPV-16 and 18, particularly, the nuclear bodies were found in all biopsies with strongly and moderately positive hybridization , and their rate of appearance decreased with weak positive and negative hybridization. No nuclear body has been found in normal epithelium of uterine cervix yet.The results from both DNA hybridization and ultrastructure research lead to the following conclusions:1. Close to 72% of all cervical cancer biopsies contain HPV-16 DNA. This result revealed that the patients with cervical carcinoma in districts of Beijing has a close relationship with HPV-16.2. Although there are related sequences of HPV-DNA in the host cell DNA, HPV particles have not been observed. The appearence of HPV -DNA related sequences in cells is suggested to be associated with features of cell ultrastructural morphology.
