高脂血清与正常血清对离体培养动脉平滑肌和斑块段细胞的影响——Ⅱ.培养细胞的蛋白质合成

【摘要】 <正> 高血脂是目前已肯定的致动脉粥样硬化(Atherosclerosis简称AS)的危险因素之一。本文的第一部分观察离体培养的动脉平滑肌细胞(SMC)经高脂血清作用后,和高脂膳食建立的AS斑块段细胞(PSC)的形态变化。第二部分的实验将分析高脂血清影响下的动脉SMC和PSC合成蛋白质功能的变化,以及这些细胞回复用正常血清培养后,其合成蛋白质功能是否有逆转正常的可能,以此探讨运用离体细胞研究AS消退机理的可能
【Abstract】 The effects of normal and hyperlipidemic rabbit serum on synthesis of protein in rabbit aortic SMC and atherosclerotic PSC were studied. 5% either normal or hyperlipidemic serum (HLS) was added to the media of 3rd to 10th passages subcultured cells for one week. Amount of DNA (D) and protein (P) was measured in each sample and P/D ratio was calculated. The results indicate that the cells, including SMC and PSC, exposed to HLS showed 69% and 123% increase in P/D ratio respectively. After the HLS treatment, cells returned to normal serum culture for another week. The P/D ratio of SMC fell to the normal level, but PSC kept in a higher protein synthesis level of 102% increase. This implies that SMC exposed to HLS and PSC synthesize protein actively, but PSC may retain their hyperactivity, even after return to normal culture environment.
