金属镍粉和氯化镍对叙利亚地鼠胚胎细胞转化的研究

【摘要】 国内外流行病学调查和实验室研究已经证明某些镍化合物在进入人体或细胞后具有潜在的致癌性。1980年对成都电冶厂341名镍冶炼工人职业危害调查时,发现工人的胸痛、咽痛、鼻干等症状和咽炎、鼻粘膜萎缩等体征均高于正常对照组并且两者间有显著性差异,且肺癌的死亡率亦较高。为此,我们用从该厂采集的金属镍粉和氯化镍对叙利亚地鼠胚胎细胞进行短期克隆转化试验,结果发现镍粉在促癌剂TPA的促进下出现了明显的形态转化克隆,转化率达6.78%。与强致癌剂MCA阳性对照组的转化率(7.23%)无显著性差异。这表明镍粉对细胞有直接的致癌作用。关于镍的致癌机理,目前还不清楚,可能与抑制RNA聚合酶有关.

【Abstract】 In a survey 1981,symptoms andsigns as chest pain,sore throat,dryness of nasal passage,laryngi-tis,nasal mucous atrophy etc.aswell as cancer mortality were incr-eased significantly among employeesof Chung-Doa smelt plant.Test ontransformation of cultured Syrianhamster embryo cell with nickel pow-der and nickel chloride collectedfrom this plant was conducted Mor-phologically transformed clones wereobserved under the combined ef-fect of nickel powder and promoterTPA.The rate of transformationwas 6.78%,comparable to that ind-uced from strong carcinogen 3-me-thylcholanthren.(7.23%) No statisti-cal significant difference was foundbetween them.The result indicatesthe carcinogenicity of nickel pow-der.However,the mechanism is stillnot clear.Probably it might besuggested that the mechanism issomething related to inhibition ofRNA polymerase.
