猪粪和玉米秸厌氧发酵两株主要产甲烷杆菌的鉴定
Identification of Two Main Methanogenic Strains Isolated from the Anaerobic Digester for Pig Dung and Corn Stalk
【摘要】 <正> 猪粪和玉米秸是我国农村沼气发酵池中常用的原料,在我们开展沼气发酵微生物研究工作中也选用了这种原料进行发酵,并分离了其中的产甲烷细菌。本文报道了从中分离到的两株主要的产甲烷杆菌的特征及对它们的鉴定。
【Abstract】 Two strains of methanogens were isolated from anaerobic digesters for pig dung and corn stalk. One strain PG 03 was non—spore forming, non—motile rods, Gram staining positive, 0.4 um in width. Length varied from 2.5um to long filaments, occurring singly or in chains. Cultures In liquid medium might have a tendency to form twisty filaments. H2+CO2 and a slight amount of formate were used as energy sources. Acetate could help the cell enrichment, but could not be utilized to produce methane. Yeast exttact, trypticase, digester fluid; rumen fluid or a vitamin mixture in mineral medium could greatly stimulate the growth. The optimum temperature for growth was 35℃—45℃; very little or no growth at 50℃. The optimum pH ranged from 6.4—7.5. According to the charcteristics of strain PC03, it could be identified as Methanobacterium jormicicum.Another strain PG25 was straight to slightly curved rods, 0.4 um wide usually 2—5 um long; sometimes in short filaments, singly or in pairs, occasionally in chainst non—spore forming and non—motile. In roll—tubes surface colonies were gray white, slightly convex, circular and translucent with entire margins. H2+CO2 and formate were used as energy sources. Methanogenesis could not occur on acetate, methanol, methylamine and trimethy—lamine. The optimum temperature for growth was 35℃—45℃. The optimum pH ranged from 6.5—7.8. It repuired the same stimulated growth substrates as those of the strain PCO3, The G+C content of the DNA was 40.3 moles% (Tm). It was also assigned as a Methanobacterium formicicum.
