三种抗人IgM免疫荧光试剂对人外周血B淋巴细胞检测的比较研究
MEASUREMENT OF B LYMPHOCYTES IN HUMAN PERIPHERAL BLOOD BY IF-LABELLED ANTI-IgM ANTIBODY
【摘要】 使用(马抗人IgM-IgG-FITC,羊抗马γ球蛋白一IgG-FITC,羊抗人IgM-F(ab)2-FITC)三种抗人IgM荧光试剂,以直接法和间接法对34例正常人外周血单个核细胞中膜表面免疫球蛋白M(SmIgM)阳性的B淋巴细胞进行了检测,均值分别为10.9%,10.7%,11.6%,均值间差异不显著。说明用抗人IgM荧光抗体检测B细胞,基本不受Fc段和方法的影响。同时,认为完整抗体直接法可简化实验步骤,较适合临床应用;而间接法较直接法敏感,在缺少特异试剂时,可参考使用。
【Abstract】 Using three different kinds of the anti-human IgM reagents (i. e. horseanti-human IgM-IgG-FITC, sheep anti-horse globulin-IgG-FITC and sheepanti-human IgM-F(ab)2 -FITC) in DIF and IIF tests, the authors examinedSmIgM+B cells of the mononuclear cell of normal human peripheral blood in 34cases, the average numbers of which were 10.9%, 11.6%, 10.7% respectively.Thus experimental procedure can be simplified by using whole IgG antibody toIgM-DIF, so it may be beneficial for clinical application. Because IIF is farmore sensitive than DIF and saves reagent, it is a methed of choice in thecondition where reagents are lacking.
