莼菜多糖的提取、分离及某些生物活性的研究
EXTRACTION, ISOLATION AND SOME BIOLOGICAL ACTIVITIES OF POLYSACCHARIDE OF BRASENIA SCHREBERI
【摘要】 人工培养法得到的莼菜Brasenia schreberi J.F.Gmel.经热水提取,氯仿-正丁醇除蛋白,透析,乙醇沉淀,沉淀干燥物再经DEAE Sephadex A-25柱层析纯化,即得莼菜多糖(简称BS)白色粉末。经气相层析和化学分析表明,BS由岩藻糖、阿拉伯糖、木糖、甘露糖、半乳糖和葡萄糖醛酸组成。红外光谱表现出明显多糖吸收峰,紫外光谱表明无核酸和蛋白质吸收峰。BS能够增加小鼠免疫器官的重量,增加腹腔巨噬细胞的吞噬功能,还有促进溶血素生成与促进~3H-TdR和~3H-UR掺入淋巴细胞核酸的作用,表明BS有较好的增加体液免疫和细胞免疫的功能。
【Abstract】 The polysaccharide (BS) was isolated from the Brasenia schreberi J.F.Gmel which grows in China. BS was further purified by DEAE Sephadex A-25. Its monosaccharide composition was identified by gas chromatography. BS was composed of L-fucose. L-arabinose, D-xylose, D-mannose, D-galactose and glucuronic acid. Intraperitoneal injection of BS in mice was found to increase the weight of spleen. The spleen index increased by 0.41 times as compared with the control. BS caused an increase in phagocytosis of macrophage by subcutaneous administration in mice, and the phagocytic index increased by 1.24 times. In addition, BS promoted the production of hemolysin by intraperitoneal injection in mice, and the HC50 increased by 0.66 times. BS stimulated the lymphocyte transformation, the incorporation of 3H-TdR and 3H-UR increased by 26 or 0.28 times respectively compared with the control.
