葡萄球菌的原生质体融合及遗传转移
PROTOPLAST FUSION AND GENETIC TRANSFER OF STAPHYLOCOCCI
【摘要】 本文报道在金黄色葡萄球菌多剂抗生素耐药菌株(22)及白色葡萄球菌(Rifr,Nalr)间进行的原生质体融合试验。在高渗缓冲培养基中,将对数生长期中、后期的培养物,以溶菌酶及溶葡萄球菌素处理,DM3培养基再生得原生质体。将二亲本葡萄球菌的原生质体混合,以CaCl2及PEG(6000)处理使发生融合及种间遗传转移。根据重组子的色素、耐药性及在琼脂糖凝胶电泳中显示的质粒带进行遗传分析,证明在金黄色葡萄球菌及白色葡萄球菌中,质粒和染色体的遗传标记均可相互转移,再次证实在金黄色葡萄球菌(22)中存在分子量为2.0Mdal的pcr质粒。
【Abstract】 The experiments of protoplast fusion between a multiple antibiotic resistant strain of S. aureus(22) and a strain of S. albus (Rifr, Nalr) are reported. Protoplast formation and regeneration were carried out by mid-late stage culture of exponential growth phase, treatment with lysozyme and lysostaphin in hypertonie buffer medium, and regeneration on DM3 medium. Protoplast fusion and interspecies genetic transfer occurred after mixing staphylococcal protoplasts, and treatment the mixture with CaCl2 and polyethylene glycol 6000. The genetic analyses were perfomed on the basis of the pigment, drug resistance, and agarosc gel elcctrophoresis. It was indicated that the mutual genetic transfer could occur not only of plasmid but of chromosome in S. aureus and S. albus. It was also demonstrated that S. aureus(22) harboured a plasmid relating to per(MW. 2.0 Mdal).
