【摘要】 本文报导了不同的培养条件对人白细胞介素2（IL2）产量的影响。结果指出在制备大剂量 IL 2时用多个供者细胞混合培养较单个供者细胞单独培养者产量高;前者若用 PHA 与多种 B 淋巴母细胞系细胞共同刺激则较单独用 PHA 刺激者其 IL 2产量又更高。在制备小样本 IL 2时将细胞预孵育24小时后再加 PHA 刺激者比新鲜细胞立即加入 PHA 刺激者所产生 IL2含量要高,有显著差异;但在无血清培基中,以上述完全相同的条件进行培养时则二者的差异无显著性。本文尚指出在评价 IL 2活性高低时,不适宜的 IL 2检测程序及条件可造成假性的 LI 2低值。由于当前无论作 IL 2研究或临床实践应用都常需要培养有关细胞以获得 IL 2,本工作似可供从事 IL₂有关实际工作者参考。更多还原

【Abstract】 Described herein is the cell cultured condition that may effect on the productof human Interleukin 2(IL2).The results showed that the pooled mononuclearcells from several blood donors produced higher titered IL2 than from single donor,and had much higher production when the pooled cells were preincubated for 4days in 10% FCS medium,followed by incubating in 1% FCS medium with both 1%PHA-M and pooled B lymphoblastoid cells.In 1% FCS medium with PHA,cellspreincubated for 24 hours produced more IL2 than the fresh cells did,however,inserum-free medium with PHA,the IL2 titer in both condition showed no signifi-cant difference.So it was suggested that for obtaining large scale of high titeredIL2 preparation,the 96-hour-preincubated pooled donor cells cultured with multi-ple stimuli would provide a satisfactory amount;whereas for a small sample IL2preparation,it might be recommended that a 24-hour-preincubated cells be culturedin 1% FCS medium with PHA for further 18-24 hours.It was indicated also thatsome unfavorable conditions of IL2 assay might cause a false low value of IL2activity.更多还原
