丙二醛对脂质体和红细胞膜作用的激光Raman光谱研究

【摘要】 本文考察丙二醛处理前后的DPPC脂质体、DPPE脂质体和红细胞膜的Raman光谱的变化.我们发现丙二醛使DPPC脂质体相变温度上升10℃且流动性增加,以丙二醛处理DPPE脂质体后其预相变消失,相变温度上升,丙二醛可交联DPPE上的氨基形成Schiff碱.人血红细胞膜与丙二醛保温后膜磷脂trans构象减少,有序性下降;膜蛋白二级结构发生明显变化,α螺旋构象减少.

【Abstract】 The Raman spectra of the DPPC, DPPE liposomes and erythrocyte membranes before and after treated by MDA have been examined. MDA increases the DPPC liposome phase transition temperature by 10℃ and decreases the fluidity. Adding MDA to DPPE liposomes results in the disappearance of pre-transition peak and the increase of phase transition temperature. It’s found that MDA crosslinks the amino groups of DPPE to form Schiff base. The comparison of the achieved high singnal-to-noise spectra of hemoglobin-free human erythrocyte membranes and the liposomes formed by the total extracted lipids from erythrocyte shows some new suggestions concerning the assignments of Raman lines of erythrocyte membrane.The of fraction of trans conformation of membrane lipids decreases and the secondary structure of membrane proteins changes greatly in the MDA treated erythrocyte membranes.
