中华眼镜蛇细胞毒素溶液构象的激光拉曼光谱研究
THE RAMAN SPECTROSCOPIC STUDIES ON THE CYTOTOXINS FROM CHINESE COBRA(NAJA NAJA ATRA)
【摘要】 由中华眼镜蛇细胞毒素CTⅡ、CTⅢ和CTⅣ水和重水溶液的拉曼光谱,按照Lippert的方法计算了它们的二级结构含量.β折叠含量为33-35%,无规卷曲为59%.后者包括大量β回折结构.根据S-S伸缩振动频率,C-C-S-S-C-C构型都属于扭曲—扭曲—扭曲式.它们的三个酪氨酸残基有两个埋藏在分子内部,一个暴露在分子表面.联系已知结构的神经毒素erabutoxin b对上述结果进行了论讨.
【Abstract】 The Raman spectra of the cytotoxins CT II-IV from Chinese Cobra in H2O andD2O have been measured and analysed. The secondary structure contents of cytotoxins were calculated with the method developed by Lippert et al., 33-35% of amino acid residues is in β-sheet and 59% in random coil or β-turn. All these toxins have gauche-gauche-gauche conformation of C-C-S-S-C-C linkage according to S-S stretching vibration frequency .Two of three tyrosyl residues of these toxins are buried, the other one is exposed.Some discussions have been made in relation to the Raman Spectra and structral data of erabutoxin b.
