YHPD及其分离组份在溶液中和CHO细胞内的吸收和荧光特征
ABSORPTION AND FLUORESCENCE PROPERTIES OF HEMATOPORPHYRIN DERIVATIVE(YHPD) AND ITS COMPONENTS IN SOLUTION AND IN CELLS
【摘要】 本文报道了YHPD及其用HPLC方法分离得到的四种主要组份在溶液中和CHO细胞内的吸收和荧光特性.YHPD分离组份d在稀溶液状态(0.5μg/ml)与其它三种组份不同,表现出较稳定的聚集态性质,YHPD及组份摄入细胞后,荧光光谱和Soret吸收峰明显红移,荧光量子产额增高.同时看到YHPD的吸收和荧光特性受环境中H~+浓度的影响
【Abstract】 The fluorescence and absorption properties of hematoporphyrin derivative (it is produced in Yang Zhou and refered to as YHPD) and its 4 main components separeted by HPLC were studied in solution and in CHO cells. The effects of solvent pH on these properties were also observed.The measured results of absorption spectra, fluoresence emision spectra, fluorescence quantum yields showe that the components a,b,c have a properties of monomeric porphyrins under 0.5μg/ml con. However, the component d differs from them, its soret absorption band not shift as the solution is diluted. The A395/A371 is smaller, and the fluorescence quantum yields is lower, which showes that the component d has more steady aggregative properties. When these components were taken by cells, the fluorescence emission spectra and absorption band were redshifted, and the fluorescence quantum yields increased. In addition to 630 nm and 700 nm fluorescence emission peak, another new one appeared at 585 nm. It is observed that H+concentration in solvent has significant effect on these properties.
