癌血清对小鼠SCM反应的特异抑制作用
THE SPECIFIC INHIBITORY EFFECT OF CANCER SERUM ON MOUSE LYMPHOCYTE SCM REACTION
【摘要】 癌血清对小鼠脾淋巴细胞SCM反应的抑制作用可用于区别健康者、非癌患者和癌症。前者RRSCM<80—85%,后者>90%。癌血清的这种抑制作用呈浓度和时间相关,无种属特异性,而仅与肿瘤的恶性与否相关。在癌症早期可出现这一抑制作用,并随癌肿的移去而消失。与Cercek经典方法或同位素参入等方法相比,本方法在易判性、可靠性、简便性方面都具优点、有理论研究和实用价值。
【Abstract】 Structuredness of Cytoplasmic Matrix(SCM),a concept developed by L.Cercek in1974,has been used to describe a certain bio-physical state of the cell.The lymphocytesstimulated by mitogens or antigens have adecrease in fluorescence polarization value(P),which is the evidence of a SCM response.Some researchers,however,thought that itwas difficult to repeat Cercek’s experiments. In 1982,we modified the Cercek’s procedureand successfully established a new system for themeasurement of SCM response.Lymphocytes witha specific density from different strains mice werestimulated and responded when the mixed lym-phocytes were incubated for 30—45min.Thusa SCM response can be induced and a decreasedSCM reaction ratio (RRscm) is given. As indicated in our experiments,we at-tempted to find out about some factors whichinhibit SCM response in the serum of cancerpatients and conditions of the inhibition usingthe SCM reaction model developed in our Lab.Mouse lymphocytes were preincubated withhuman sera from healthy persons and patientswith various diseases other than cancer.Theresults indicated that the sera from healthydonors (26/26) give a RRscm=75.5±3.1%,from non-malignant diseased patient (33/34),76.3±5.7% and from cancer patients(48/49),98.3±4.6%.The inhibition of SCM responseis related to the concentration of cancer serumand the preincubation time,but not related tomouse species.The inhibition became apparent4 hrs after implantation with Ehrlich ascitescells and disappeared after surgical removal ofa cancer for 2 weeks or more.These resultsstrongly suggest that the mouse SCM model candistinguish cancer from non-malignant diseases,and may be very promissing in cancer diagnosis. Compared with Cercek’s SCM responsesystem,our model has many advantages.Theimportant things are that the amount of bloodneeded is greatly reduced (from 15 ml to 2ml) and better reproducibility are achieved. Since the early 1970’s,there have been manyreports on an inhibitor in tumour serum whichinhibits [3~H] TdR incorporation in lymphocytetransformation reaction and PFC.However,thiskind of inhibition is observed not only withtumour serum,but also with the sera of manyother non-malignant patients,so these tests havenot been successfully used in cancer differencia-tion.But the method reported here is specific,The serum factor in cancer which specificallyinhibits SCM response may belong to an earlyimmunological reaction inhibitor.
