高效液相色谱法测定血清去甲肾上腺素及肾上腺素
SERUM NORADRENALINE AND ADRENALINE QUANTITATION BY REVERSED-PHASE HIGH PERFORMANCE LIQUID CHROMATOGRAPHY
【摘要】 本文报道用国产高效液相色谱仪联电化学检测器,反相内标法测定血清去甲肾上腺素(NA)及肾上腺素(AD)。研究了不同流动相条件下对血清NA及AD容量因子(K′)及响应值的影响,从而选择最佳测定条件。血清NA及AD浓度在0.2~10ng/ml内与色谱峰面积比值的线性关系良好(r>0.99);不同浓度的血清NA和AD绝对回收率及方法回收率分别为74.4~89.9%及95.1~107.6%;不同浓度的日内、日间变异系数分别为5.0~9.7%及5.3~11.9%。测定10名正常人及4例嗜铬细胞瘤患者的术前血清NA和AD值,二者有显著差异。提示本法对诊断嗜铬细胞瘤有一定的临床价值。
【Abstract】 A sensitive and specific procedure for quantitation of serum nor-adrenaline (NA) and adrenaline (AD)using a domestically produced high performance liquid chromatograph in combination witn an electrochemical detector (HPLC-EC) is described. The capacity factor (K) and signal value of serum NA and AD affected by different mobile phases were studied. 3, 4-Dihydroxybenzylamine (DHBA) was used as an internal standard. Linear calibration curves for serum NA and AD were measured within a rango of 0.2-10.0ng/ml. The average correlation coefficient was over 0.99. In three different concentrations, the absolute and relative recovery rate of serum NA and AD were between 74.4-89.9% and 95.1-107.6%, respectively. The within a day, and day to day 0V were between 5.0-9.7% and 5.8-11.9%, respectively. The results of serum NA and AD in 10 healthy subjects and proven preoperative, cases of pheochromocytoma, were significantly different, with p<0.05. It suggests that this procedure is a useful tool in detecting serum concentrations of NA and AD, which is not only valuable for diagnosis of pheochromocytoma, but also in research of many relevant patho-physiological conditions.
