【摘要】 用白细胞介素2(interteukin 2,IL2)依赖细胞株(Cytotoxic T-lymphocyte line,CTLL)检测IL2并滴定标本中IL2的活性单位,同时观察诱导IL2产生的条件和影响因素。结果表明CTLL对IL2绝对依赖,对植物血凝素(PHA)和伴刀豆球蛋白A(Con A)无反应。CTLL的增殖与IL2的量成正比。Con A刺激大鼠脾细胞的培养上清液(又称大鼠因子,RF)中有可透析的小分子抑制因子存在,如不除去会影响IL2活性单位的滴定。大鼠IL2的诱生受下列因素影响:①个体差异;②Con A浓度,IL2的产量随Con A浓度的增加(3μg～50μg/ml)而增加;③牛血清白蛋白(BSA)可以替代小牛血清(FCS),但IL2的产生受到影响。更多还原

【Abstract】 We report the development of a sensitive mioroassay for interleukin 2 (IL 2) activity. The IL 2 assay is based on the dependance of cultured murine cytotoxic T-lymphocyte line (CTLL) on IL 2. The results indicated that growth of OTLL cells were dependent on IL 2 in a dose dependent manner and OTLL cells did not respond to phytohemagglutinin and concavanallin (Con A) in the absence of IL 2. The culture supernatants derived from Oon A-stimulated rat spleen cells (Rat Factor, EF) had 0.20 unit/ml of IL 2 activity after dialysis. Further investigations demonstrated that (1) there are differences in the ability to produce IL 2 among different individuals; (2) the quantity of IL 2 produced by Con A stimulation is dependent on the dosage of Con A; (3) RPMI 1640 medium supplemented with bovine serum albumin also could be used in IL 2 production.更多还原
