影响纤维素酶水解木屑的若干因子的研究
STUDY ON THE EFFECTS OF SOME FACTORS IN THE HYDROLYSIS OF SAWDUST BY CELLULASE
【摘要】 木霉201、79、SA3和1096产生的纤维素酶对天然木屑的水解能力都极其低微,但当木屑经过适当的预处理以后,酶的水解能力明显地提高。X2-85菌纤维素酶在45℃保温24h,酶活损失15%左右。用三种底物考察酶用量与酶解率之间的关系:酶浓度增加100%,木纸浆的酶解增长率为120%,脱脂棉的酶解增长率为31%,酸处理木粉的酶解增长率为2%。差别很悬殊,说明提高酶解率的主要途径,是把对酶亲和力小的木屑转变为对酶亲和力大的底物。
【Abstract】 The ability of cellulase produced by strains 201,79, SA3 and 1096 for attecking sawdusts were low,while that for the pretreated sawdust were hight. The activity of the X2-85 enzyms lost 15% after incubated in 45℃ for 24h. When the enzyme incubated with three different substrates at the same time, the increase of enzyme concentration for 100% resulted an increase of hydrolytic rate of wood pulp to 120%, but that for the absorbent cotton and acid treated wood powder only 31% and 2% were observed respectively. The great difference among the three substrates in hydrolytic rate suggest that the primary ways of increasing the hydrolysis of sawdust is to seek the methods for changing the sawdust substrate from small affinity to great affinity for enzymes.
