一组抗胸腺细胞单克隆抗体的制备与鉴定——Ⅱ.生物学特性和临床的初步应用

【摘要】 本文主要报道一组抗胸腺细胞单克隆抗体的生物学特性及其在临床上的应用。HIT4和 HIT1 在补体存在下有明显的细胞毒作用。其它单抗结合补体能力很弱。HIT1-3抗原出现在激活T细胞表面,不出现在休止外周T细胞上。HIT4和HIT6-1.2与休止和激活T细胞都不发生反应。HIT2和 HIT3单抗共同识别一个 43KD的抗原决定簇,而 HIT4则独立识别一个 35KD的抗原。脐血单个核细胞中 HIT1-3和OKT10阳性率明显高于正常外周血,但Tac 阳性率较低,提示脐血中存在大量不成熟T细胞。通过研究这一种单抗的生物学特征,我们认为人类T细胞分化过程可能比 Rein-herz提出的模式更为复杂。我们还联合应用 HIT1-3和 HIT4及其它标准单抗对白血病细胞表现型进行分析和研究。

【Abstract】 A group of monoclonal antibodies (McAb) against human thymocyte differentiation antigen, named HIT1, HIT2, HITS, HIT4, HIT6-1 and HIT6-2, were produced and reported. HIT1-3 are OKT10-like antibodies and HIT6-1, 2 are OKT6-like. HIT4 seems to be a new McAb different from HIT1-3 and HIT6-1,2. This paper describes their biological properties and application on clinical leukemia classification.HIT4 (IgM) and HITl (IgG3) are able to cause target cells lyses in the presence of complement. Other McAbs are IgG1 with a weak capacity of complement fixation. Quantitative absorption test showed that the antigens recognized by HIT1-6 McAbs are not present in human brain. The pattern of HIT1-3 antigen on the surface of mitogen activated T-cell are very similar to OKT10. However, HIT6-1,2 and HIT4 do not express on activated T-cell. HIT2 and HITS immunoprecipitate a polypeptide of 43KD, HIT4 antibody immunoprecipitates a 35KD molecule. Combination of HIT1-3 and HIT4 McAb is good for analysis of leukemia surface markers. They are useful for classification of leukemia and study of T cell differentiation.We also found cord blood has higher percentage of HIT1-3+ and OKT10+ cells, suggesting immaturity of T-cell in cord blood. Besides Reinherz model, alternative pathway of T-lymphocyte differentiation has been discussed. Antigen modulation was not induced by these MoAbs.
