凹叶景天叶片离体培养的形态发生
MORPHOGENESIS OF BLADE CULTURE OF Sedum emarginatum IN VITRO
【摘要】 凹叶景天(Sedum emarginatum)叶外植体于MS+0.2mg/l NAA+2mg/l BA固体培养基上诱导培养,形成愈伤组织,并进一步产生大量的芽。经石蜡切片观察,愈伤组织形成时,细胞核的分裂类型多样,都属无丝分裂,未观察到核的有丝分裂。据细胞组织学断定,芽的发生途径为不定芽的途径。将已产生丛生芽的愈伤组织切块转接到MS基本培养基上后,芽体伸长、健壮。切取伸长的苗,再转接到1/2 MS(或MS)培养基上,经2—3周的培养,则在苗茎的切口处产生白嫩的不定根,形成完整植株。
【Abstract】 Leaf explants of Sedum emarginatum were cultured on MS medium supplemented with NAA 0.2mg/l and BA 2.0mg/l, 10 days after callus formation was induced, then many buds were differentiated from calluses. Histocytological observations on buds formation showed that the buds differentiated were adventi- tious buds. After calluses with clustered buds were transplanted onto hormone- free MS medium, the buds elongated and grew with vigor. The elongated stem of shoot was cut off and then transferred onto rooting MS medium. About 2-3 weeks after white and tender adventitious roots were produced from the cut end of stem and complet plantlets formed. The caryological observation on callus paraffin sec- tion showed that in course of callus formation there were three types of nuclear division modes in cell division process, but all these were of amitosis.
