液氮冻结法保藏毛霉目菌种的效果

【摘要】 本文报告了用液氮冻结法保藏毛霉目菌种的效果。对14属26种32株进行了液氮保藏试验,其结果慢速冻结孢子成活率高于快速冻结。用10%的甘油、10%的二甲基亚砜和蒸馏水作保护剂制成的孢子悬液,经慢速冻结后储存在液氮罐气态中二年,以甘油作保护剂的孢子成活率为90%,二甲基亚砜的为85%,蒸馏水的为79%。并检测了某些菌株的反丁烯二酸、蛋白酶、α-半乳糖苷酶、脂肪酶、果胶酶、葡萄糖苷酶等生理活性,结果冻结后的活性与冻结前相比,无明显差异。

【Abstract】 In a previous paper,We published an evaluation about the efficacy of mineral oil sealing,lyophilization and wheat barn methods for preservation of Mucorales cultures.In this investigation,we observed the response of Mucorales strains subjected to ultra-low temperature freezing,storage and thawing.Thirty two strains from 14 genera and 26 species of Mucorales cultures are all viable after storage in liquid nitrogen refrigerator for 2 years.The average sur- vival rate of spores protected separately with 10% glycerol,10% DMSO and distilled water was 90% 85% and 79% respectively. From the point of view of cooling rate,slow freezing,frozoen at a controlled rate of l℃/min.,gave higher survival rate than rapid freezing for which the sample was put into liquid nitrogen refrigerator immediately. Glycerol is the one of the best protective additive when it was compared with DMSO and distilled water used in ultra-low temperature freezing for preservation Mucorales cultures. The ability for fumaric acid formation and the activities of proteinase,lipase,α-galactosi- dase,pectinase and glucosidase of the examined strains retained the original level after freezing and thawing.
