家兔体外受精研究

【摘要】 研究获得了我国第一批哺乳动物“试管”后代。 利用供体母兔输卵管中采集的未受精卵,和获能母兔子宫中回收的获能精子进行体外受精。以获能母兔输卵管中采集体内受精卵作对照。结果表明,体外受精和体内受精兔卵的体外发育率分别为76.8%(43/56)和82.3%(14/17)。将体外发育的胚胎分别移植至经同期化处理的青紫兰母兔受体,从体内受精胚胎获得2只仔兔,产仔率为14.3%(2/14),从体外受精胚胎获得5只仔兔,产仔率为18.5%(5/27)。

【Abstract】 The objective was to obtain in vitro fertilization, early cleavage and normal offspring of rabbit ova. Ovum donors, New Zealand White does, were injected with PMSG or FSH and HCG to induce superovulation for recovery of unfertilized ova. Thirteen hours after treatment with HCG ova were recovered from oviducts and then transferred into a suspension of spermatozoa recovered from the uterine horns of capacitator rabbits which had been mated 18 hr. before by two fertile bucks. Ova were incubated with capacitated spermatozoa for six hours under a moist 5% CO2, 8% O2 and balance N2 atmosphere at 38℃. Then the ova were removed from the suspension and cultured in 10% serum solution at 38℃ through early cleavage stages. Cleavage rates for in vitro and in vivo (capacitator’s ova) fertilized ova were 76.8% (43/56) and 82.3% (14/17), respectively. The embryos were transferred into the oviducts of 5 Chinchilla rabbits that had been injected with HCG at the same time as the donors. One of the recipients died on fourth day after transfer. Fourteen embryos from in vivo fertilized ova transferred into two recipients resulted in two live young. Twenty seven embryos from in vitro fertilized ova transferred into two recipients resulted in five living healthy offspring. The normal development rates of in vivo and in vitro fertilized ova were 14.3% and 18.5%, respectively.
