水飞蓟素琥珀酸钠和熊果酸苯二甲酸半酯钠对原代培养大鼠肝细胞作用的研究

【摘要】 木文利用大鼠肝细胞原代培养方法研究了水飞蓟素琥珀酸钠和熊果酸苯二甲酸半酯钠对正常肝细胞及四氯化碳引起的肝细胞损伤的作用。观察了二药对肝细胞培养液中的GPT、GOT和LDH活性的影响,并作了肝细胞超微结构的电镜观察。证明水飞蓟素琥珀酸钠是特异的GPT直接抑制剂;熊果酸苯二甲酸半酯钠是GPT和LDH的直接抑制剂。二药在所试浓度均未表现出保护肝细胞、减轻四氯化碳损伤的作用。熊果酸苯二甲酸半酯钠在高浓度(1.0mg/ml)时还呈现对肝细胞的毒性怍用。文中对实验方法与结果作了必要的讨论。

【Abstract】 The actions of silybin disodium hemisuccinate and mono-(β-3-ursolic acid) phthalate disodium on the primary cultured normal rat hepatocytes and hepa-tocytes with carbon tetrachloride-induced injury were examined. The activities of GPT, GOT and LDH in the hepatocyte culture medium were determined, and the ultrastructure of hepatocytes was examined by electron microscope. The results showed that the silybin disodjum hemisuccinate was a direct GPT inhibitor, the mono-(β-ursolic acid) phthalate disodium was both a GPT inhibitor and a LDH inhibitor. These two drugs, at the concentrations used, could not protect the hepatocytes against the toxic effect of carbon tetrachloride. At high concentration (1000μg/ml) , mono- (3-3-ursolic acid) phthalate disodium showed cytotoxicity itself. The methodology and the significance of the results have been briefly discussed.
