用去HGH混合正常马血清制备HGH RIA质控样品的初步探讨

【摘要】 作者采用亲和层析法处理混合正常马血清,制得去生长激素的血清,并用以配制人生长激素放射免疫测定的质控样品。经国内22个单位临床试用半年,结果证明达到质控要求。对比试验表明:单一正常人血清经同样处理,人生长激素(HGH)不能除尽;混合人血清者反而略升高,其原因尚待探讨。

【Abstract】 Using affinity chromatography, we pre-pared HGH-immunoreactivity deprivedpooled normal horse sera. These sera wereused to prepare external quality assessment(EGA) samples, EQAs of 0,2, 10 and 30ng/ml, which have been distributed to 22institutes and used in HGH RIA for sixmonths with the same method and thesame HGH kits produced in this centre.Data of EQA determinations of all assaysfrom these institutes were collected andanalyzed by computation.The results showedthat the mean±SD for EQA1, EQA2 andEQA3 were 2.13±0.25, 10.33±0.79 and30.81±4.04ng/ml (n=254), respectivelycompared to the target values of 1.85±0. 14,10. 13±0.82 and 31.27±1. 17(n=11). Single and pooled human normal serawere also tested. The rusults showed thatthe human growth hormone immunoreac-tivity could not completely be eliminated(for single human serum)or even increased(for mixed normal human sera) with thesame affinity chromatography used. Theexplanation for the results of the mixedhuman sera is not clear and needs to befurther studied.
