马桑内酯所致癫痫大鼠的脑组织化学研究

【摘要】 以微量马桑内酯注入大鼠大脑左侧皮质运动区,引起急性局灶性癫痫。用组化技术显示AcPase和ATPase的变化,并用定量图象分析仪进行分析。结果显示:大脑皮质运动区和海马的锥体细胞、小脑蒲肯野氏细胞的AcPase活性增强;ATPase定位于星形胶质细胞和毛细血管,大脑皮质运动区和海马的酶活性增强,小脑未见显著性变化。

【Abstract】 Einghteen adult male Wistar rats were di-vided into two groups.The experimental groupwas injected in the loft motor cortex with 20 μgcoriarla lactone,and was induced to acutelocal seizures.The control group was injectedwith saline. The motor cortex of cerebrum,the cer-ebellum,and hippocampus were studied withthe enzyme histochemical tests for lysosomalacid phosphatase and Mg++ adenosine tri-phosphatase.The sections were analysedwith Quantimet 720 Image Analyser. In the experimental group,a stage ofgeneral increase in lysosomal AcPase activi-ty was detected in the pyramidal neurons ofmotor cortex,pyramidal layer of hippocampus and Purkinje cells of cerebellum.In thebrain of the control group,prominent activi-ty of Mg++ ATPase was found in the astro-cytes and capillaries.In epileptic rats Mg++ATPase showed a marked increase in astro-cytes of motor cortex and hoppocampus.Thecerebellum did not reveal changes.It is sug-gested that the lesions induced by localepilepsy influence not only the local area ofthe brain,but the more extensive area suchas hippocampus.The results showed that inepileptic rats the energy metabolism increas-ed in astrocytes besides the change of-neurons.
