山莨菪碱对兔呼吸窘迫综合征肺泡巨噬细胞糖皮质激素受体的影响

【摘要】 用山莨菪碱(654-2)防治油酸引起的兔呼吸窘迫综合征,观察分析了654-2对肺泡巨噬细胞糖皮质激素受体和支气管肺泡灌洗液中各种细胞数量、白蛋白含量及酸性磷酸酶活力的影响。结果表明,654-2可保护肺泡巨噬细胞的糖皮质激素受体,减少白细胞扣押在肺,抑制溶酶体酶释放,减轻呼吸膜通透性的变化,最终减轻肺损伤。

【Abstract】 The effect of anisodamine on glucocorticoid receptor(GCR)of pulmonary alveolar macrophages(PAM)in oleic-acid(OA)-induced respiratory distress syndrome(RDS)of rabbit disease model was observed to investigate the mechanism of anisodamine therapy.Rabbits were randomly derided into OA group and anisodamine-treated group.Anisodamine was administered 8mg/kg bolus and 4mg/ kg/h which was initiated 15 min before OA injection and lasted for 4 hrs.Results showed that anisodamine attenuated almost every disorders induced by OA.The GCR content in PAM after anisodamine treatment remained to be 12075 ±633 binding sites/cell as compared with 6442±841 binding sites/cell in the untreated group.The leukopenia occurred at 5 min after OA injection was blocked,and a neutrophilia at 240 min was presented after anisodamine pretreatment,which was aceompanied with a markedly reduced PMNs in BALF and pulmonary tissue.Furthermore,the increase of BAL erythrocytes,BAL albumin content and BAL ACP activity induced by OA was declined markedly.It is evident that the protective effect of anisodamine on GCR of PAM might play an important role in drug therapy of OA induced RDS.
