内毒素休克狗血浆Ⅷ因子相关抗原的动态变化机制
PLASMA FACTOR Ⅷ RELATED ANTIGEN IN EXPERIMENTAL ENDOTOXIC SHOCK
【摘要】 为了观察内毒素休克狗血浆Ⅷ因子相关抗原(ⅧR:Ag)的动态变化,制备了兔抗狗ⅦR:Ag 血清。结果发现:内毒素休克时血浆ⅦR:Ag 明显升高,其升高程度与休克动物的预后有关,可作为判断休克预后的一个参考指标。当血小板在体外由内毒素刺激聚集时,血浆ⅧR:Ag 含量则降低。综合体内、外实验结果,对内毒素休克时血浆ⅧR:Ag 升高的机制进行了探讨。
【Abstract】 Alterations of plasma ⅧR:Ag and its mechanisms in dog experimental endotoxic shock were studied.Seventeen adult mongrel dogs were randomly divided into two groups:Experimental group(N=12)was given a bolus injection of disintegrated E.coli(1011 organisms/kg)and an equivalent normal saline as control (N=5).Mean systemic arterial pressure,plasma ⅧR:Ag,platelet aggregation rate,platelet count and total count of white blood cells were monitored for 360 minutes. The results showed:(1)Plasma ⅧR:Ag increased significantly in dogs during endotoxic shock and at the end of 120 min,plasma ⅧR:Ag was recovered in some of them and progressively increased in others,which was well correlated with the prognosis and the convalescence of BP;(2)when platelets Were treated by endotoxin in vitro,plasma ⅧR:Ag decreased significantly.It is indicated that:The increase of plasma ⅧR:Ag in endotoxic shock may be due to a more amount of ⅧR:Ag released from damaged endothelial cells and stimulated platelets than that consumed during adhesion and aggregation of platelets;and plasma Ⅷ R:Ag level may be used as an indicator for evaluation of the prognosis of shock.
