需NADP~+与NAD~+的异柠檬酸脱氢酶和己糖激酶的组化方法学探讨
HISTOCYTOCHEMICAL METHODS ON NADP~+-AND NAD~+-DEPENDENTISOCITRATE DEHYDROGENASE AND HEXOKINSE
【摘要】 本文用50只健康成年大鼠的鼻咽上皮及其周围组织,研究了三羧酸循环中关键酶需辅酶Ⅱ(NADP~+)与辅酶Ⅰ(NAD~+)的异柠檬酸脱氢酶(T-IDH 与 DH-IDH)和糖酵解关键酶己糖激酶(HK)的组化方法。结果表明,采用本改良法显示 T-IDH、D-IDH 和 HK,比不含酚嗪硫酸甲酯(PMS)的 Bancroft 法和 Meijer 法可靠,各种酶的活性较 Reider 法和 Lawrence 法显示的为强。文中讨论了影响需辅酶脱氢酶组化反应的几个重要因素。
【Abstract】 The histochemical methods of aerobic oxidative key enzyme isocitratedehydrogenase,including NADP~+-and NAD~+-dependent isocitrate dehydrogenase(T-IDH and D-IDH)and of glycolytic key enzyme hexokinase(HK)were studied in thenasopharyngeal epithelia and around tissues of 50 normal rats.It was shown thatthe results of the modified methods of T-IDH,D-IDH and HK in three types of thenasopharyngeal epithelia were more reliable than that of Bancroft’s and meijer’smedia(without phenazine methosulfate),the activity of the three enzymes demonstratedwith modified media was stronger than that with Reider’s and Lawrence’s.Theimportance of several factors which affect the histochemical reaction of coenzyme--dependent dehydrogenases was also discussed.
