泰必利中间体-2-甲氧基-5-甲磺酰基苯甲酸的新合成法
A novel synthesis of 2-methoxy-5-methylsulfonyl benzoic acid——An intermediate for Tiapride
【摘要】 本文叙述文题化合物(Ⅱ)的合成方法改进。2-甲氧基苯甲酸经氯磺化反应生成2-甲氧基-5-氯磺酰基苯甲酸(Ⅲ),Ⅲ用亚硫酸钠在碳酸氢钠存在下进行还原反应,得2-甲氧基-5-亚磺酸苯甲酸二钠盐(Ⅳ),Ⅳ直接用硫酸二甲酯甲基化、酸析而得产物Ⅱ,收率以Ⅲ计约70%。
【Abstract】 A novel synthesis of title compound (Ⅲ) is described. 2-methoxy-5-chlorosulfo-nyl benzoic acid (Ⅲ) was prepared by chlorosulfonation of 2-methoxybenzoic acid andthen reduced with sodium sulfite in the presence of sodium bicarbonate to give disodium2-methoxy-5-sulfinate benzoate (Ⅳ) which, without separation, was followed by me-thylation with dimethyl sulfate and finally acidification to produce Ⅱ in a yield of about70% from Ⅲ.
