用甘蓝夜蛾核型多角体病毒感染豚鼠
INFECTION OF GUINEA PIGS BY NUCLEAR POLYHEDROSIS VIRUS OF MAMESTRA BRASSICAE
【摘要】 用纯化的甘蓝夜蛾核型多角体病毒悬液,按4×10~6PIB/克体重剂量,经腹腔接种豚鼠,在第6天和第16天解剖取胰脏,超薄切片电镜观察,可在胰脏细胞内发现多角体病毒;用豚鼠肝、脾和胰等脏器匀浆液饲养棉铃虫幼虫,被感染虫患典型病毒病,死虫率6天解剖的豚鼠为70.83%,16天解剖者为91.3%; 病死虫脓计在扫描电镜下发现典型多角体.据此可确定甘蓝夜蛾核型多角体病毒在豚鼠脏器内滞留16天.至于豚鼠脏器细胞是否复制该病毒问题,尚需进一步研究.
【Abstract】 Purified suspension of nuclear polyhedrosis virus of Mamestra brassicae was inoculated in the abdominal cavity of guinea pigs at a dosage of 4×106 PIB per gram body weight. 6 and 16 days after inoculation, pancreas was taken out for electron microscope examination. Strains of the polyhedrosis virus were found in the cells of pancreas sixteen days after inoculation. The suspensions of the liver, spleen and pancreas of the inoculated guinea pigs were prepared to rear the larvae of cotton ballworms ( Heliothis armigera H. ) , and the infected larvae showed typical symptoms of the virus disease. The mortality of the infected larvae was either 70.83% for the anatomi cal treatment six days after inoculation, or 91. 3% for the anatomical treatment sixteen days after inoculation. Typical polyhedras were also found in the dead bodies of the larvae by electron microscope examination. Therefore, it can be ascertianed that nuclear polyhedrosis virus of Mamestra brassicae can survive in the cells of guinea pig organs for sixteen days. Virus duplication in the guinea pig cells is also discussed in this paper. However, further research work should be done on the duplication of this virus in the cells of mammals.
