美洲商陆丝裂原(PWM)诱导淋巴细胞和脂多糖(LPS)诱导淋巴细胞的辐射敏感性

【摘要】 应用人血在体外培养,分别用PWM和LPS诱导淋巴细胞,接受各种剂量60Coγ线照射,两种细胞按各种组合混合再培养,并设有各种单项培养作对照,以3H-TdR掺入反映淋巴细胞的增殖转化。实验结果说明PWM细胞能够激活LPS细胞转化,当一种细胞受照后单独培养或与另一种正常细胞混合培养,其放射性掺入即下降,并随剂量增加而愈益明显、下降趋势均呈现负相关,并分别得到直线回归方程,差异显著性比较说明PWM细胞受照影响更严重。当PWM细胞接受1Gy以上剂量后与正常LPS细胞混合培养即失去激活作用,LPS细胞受照1—2 Gy仍然可被激活,以上提示电离辐射导致机体免疫缺陷,其中PWM诱导的T辅助细胞受损是较重要的。

【Abstract】 Human whole blood were incubated in vitro. Lymphocytes were in-duced by mitogen PWM and LPS respectively. After exposure to 60Coγ-rays at variousdoses, the cells were incubated together in different combination. Lymphocyte inducedby one kind of mitogen was incubated as control. The rate of lymphocyte pro1iferationand transformation was determinated by 3H-TdR incorporation. This study demonstra-ted that PWM-induced lymphocyte was able to enhance the stimulating effect of LPSon B lymphocyte. Lymphocytes induced by one kind of mitogen were exposed to 1-40Gy r-rays, and the cells were incubated alone or with the lymphocyte induced byanother mitogen, thus, the 3H-TdR incorporation became much lower than that in con-trol. The result showed that the decrease in incorporation of 3H-TdR was dose depen-dent and it had a negative correlation. The relationship between the logarithm of ra-diation dosage and the radioactivity counts for 3H-TdR incorporation was expressedby the linear regression equations. By T-test, it was shown that PWM-induced lym-phocyte was damaged more markedly than LPS-induced lymphocyte by irradiation. Thestimulating effect of the PWM-induced lymphocyte disappeared after 1 Gy irradiation.but the LPS-induced lymphocyte after 1-2 Gy irradiation was still stimulated by thePWM lymphocyte. It demonstrated ionizing radiation caused defect of immunity inorganism, the damage of the PWM-induced T helper lymphocyte was more importantin maintaining the function of immunity.
