抗Le~a和抗Le~b血清的研制
PREPARATION OF ANTI-Le~a AND ANTI-Le~b SERA
【摘要】 利用进口抗Le~a、Le~b血清筛选OLe(a+b-)和OLe(a-b+)型人,测定其唾液中Le~a和Le~b型物质含量,择其型物质含量高者唾液,用家兔和山羊免疫,合格后采全血,分离血清,用O、A、B型红细胞吸收、除去种属及α和β等凝集素。再用O型Le非相应型红细胞吸收,精制出含不完全抗体的抗Le~a和抗Le~b血清。该血清对Le相应型红细胞均产生明显凝集反应,而不发生非特异的交叉凝集反应。经过盲测鉴定,17份红细胞的Lewis型测定完全准确。制备的抗Le~a和抗Le~b血清在效价和特异性方面达到了引进的同种血清水平,填补了我国抗Le~a、抗Le~b血清制造的空白。
【Abstract】 Anti-Le~a and anti-Le~b sera were prepared by immunizing rabbits or goats with Le~a and Le~b antigens in human saliva. Firstly, Lewis typing of group O donors were carried out with anti-Le~a and anti-Le~b Sera("Ortho" and "Dade"). And then saliva samples with higher level of Le~a or Le~b substance were collected. Le~a and Le~b antigen were prepared by the method of absolute alcohol sedimentation and kept freeze-dried. Rabbits were immunized with 5rag (10mg for goats) lyophilized Lewis substance mixed with freund adjuvants intra vinously and subcutaneously for seven times at one week interval being 0.5mg more than the preceding one. The titers of Anti-Le~a and Anti-Le~b sera were examined 10 days after the last injection. The rabbit was bled once the titer of the antiserareached 256~512.The raw antisera were absorbed with ABO and antithetic Lewis antigens and then absorbed with papain treated red cells. The mono-specific anti-Le~a and anti-Le~b sera were obtained after repeated absorption for 7~11 times. The titer was 4~16.The immuned anti-Le~a and anti Le~a antibodies were incomplete antibodies. Therefore, the papain treated red cells suspended in normal saliva must be used as indicator cells,
