【Abstract】 In this paper a method for preparing DNA from a small number (10 to 40) of hand-iso-lated macronuclei of two hypotrichous cilliates for electron microscopy is discribed. This me-thod is especially advantageous in case in which mass isolation of nuclei is difficult or largenumbers of nuclei of a particular stage of cell cycle or development cannot be obtained, andthe method involves minimal manipulations compared with routine nucleic acid extractions andis extremely rapid. Although the method was devised for macronuclei of hypotrichous ciliates,it may also be useful for analysis of the DNA of other large eucaryotic nuclei.更多还原
