正常家兔小肠粘膜受体蛋白在1,25-二羟维生素D3测定中的应用
THE APPLICATION OF RECEPTOR FROM NORMAL RABBIT INTESTINAL MUCOSA FOR 1,25-DIHYDROXYVITAMIN D3 ASSAY
【摘要】 本实验采用国产家兔(Oryctolagus cuniculus domestica中国本兔),用差速离心法从其小肠粘膜上提取出1,25-(OH)2D3受体蛋白。将此受体蛋白与来自佝偻病小鸡十二指肠粘膜的受体蛋白相对照,进行放射性竞争结合试验,并绘制出标准曲线。结果表明,兔小肠粘膜1,25-(OH)2D3受体蛋白与传统采用的佝偻病小鸡受体蛋白一样,可用于人血清1,25-(OH)2D3的放射性竞争结合测定,正常值为37±12pg/ml,与国外报道的结果一致。兔的受体蛋白Kd=0.054nmol/l,最大结合量为74fmol/mg蛋白质。因此,国产的正常家兔可作为受体蛋白的廉价来源用于1,25-(OH)2D3的研究和测定。
【Abstract】 In the present paper, a specific receptor protein for1, 25-dihydroxyvitamin D3 (1, 25-(OH)2D3) was prepared from the smallintestine of vitamin D-sufficient rabbits (the Chinese white rabbits,Oryctolagus Cuniculus Domestica) by differential centrifugation. Thisreceptor was used in the competitive protein binding radioassay andcompared with the receptor from rachitic cockerels. Standard curves of1, 25-(OH)2D3 binding assay were drawn. As the receptor from theintestinal mucosa of rachitic cockerels did, the receptor from normalrabbits was suitable to perform the competitive receptor bindingradioassay for measuring human serum concentration of 1, 25-(OH)2D3.The normal value (37±12 pg/ml) was similar to that in the reportsfrom other countries. The dissociation constant of rabbit recep-tor 1, 25-(OH)2D3 complexes was 0.054 nmol/L and maximum bindingcapacity was 74 fmol/mg cytosol protein for 1, 25-(OH)2D3. It is there-fore convenient to use Chinese white rabbits as a plentiful, inexpen-sive source of binding protein for 1, 25-(OH)2D3 assay.
