家蚕质型多角体病毒双链核糖核酸分子杂交的研究

【摘要】 为了研究家蚕质型多角体病毒(CPV)的致病机理及其早期检测,我们建立了该病毒双链核糖核酸(dsRNA)分子杂交技术.dsRNA分子杂交的关键在于其解链及使解链后的单链RNA(ssRNA)与其互补DNA杂交.我们采用90%的二甲基亚砜或8M的尿素使CPV—dsRNA解链.并合成了它的cDNA探针.用此探针完成了硝酸纤维膜上的CPV核酸斑点及基因组片段转移分子杂交的研究.

【Abstract】 In order to study the mechanism of the pathogenesis of CPV and diagnosing the disease early, we have reserached the technique of viral double-stranded RNA (dsRNA) molecular hybridization. The key procedures of the dsRNA hybridization are melting the dsRNA into the single-stranded RNA (ssRNA ) , and hybridizing the ssRNA with cDNA probe. The CPV-dsRNA were denatured and melted into ssRNA with 90% dimethyl sulfoxide dr 8M urea. Then cDNA probe of CPV RNA were synthesized by the method of reverse transcriting and finished the research of the CPV RNA molecular hybridization of blots and genome segments transfering.
