二氢农吉利碱对人体肝癌细胞系及小鼠艾氏腹水癌细胞的影响

【摘要】 二氢农吉利碱可降低人体肝癌细胞系(BEL-7402)细胞的生长和有丝分裂指数,增加其死亡率,对小鼠艾氏腹水癌细胞数目的增长也有抑制作用。农吉利碱的烯丙酯结构,不是农吉利碱抗癌活性的主要部位。改变双稠吡咯环1、2位双键,生成的二氢农吉利碱仍具有抗癌性,而对肝脏的毒性作用较小。二氢农吉利碱LD50为1042 mg/kg(i.p.)。

【Abstract】 The growth, the survival rate and the mitotic index of the line of human liver cancer cells (BEL-7402) may be significantly reduced by the action of dihydromonocrotaline.Dihydromonocrotaline is capable of killing the human liver cancer cells directly and inhibiting the growth of Ehrlich’s ascitie cells of mice as well.The chemical structure of the allyl ester of monocrotaline is not the primary and active antican- cerous position of monocrotaline.The formation of dihydromonocrotaline after the hydrogenation of the double bond the 1,2 position of pyrrolizidine of monocrotaline still maintains the anti-cancerous effect as well and the toxicity of dihydromonocrotaline on the livers of mice is smaller than that of the monocrotaline.The acute LD50 of dihydromonocrotaline in mice is found to be 1042 mg/kg (i. p.).
