上消化道癌瘤患者组织中谷胱甘肽硫转移酶和γ—谷氨酰转肽酶活性的测定
MEASUREMENT OF HUMAN TISSUE γ-GLUTAMYL TRANSPEPTIDASE AND GLUTATHIONE S-TRANSFERASE IN PATIENTS WITH MALIGANT TUMORS OF DIGESTIVE TRACT
【摘要】 目前,食管癌、贲门癌的病因尚未完全阐明,为探讨化学致癌剂对其发病的影响,测定了13例食管癌、贲门癌患者正常组织与病理组织中γ—GT和GSH—S—转移酶的活性。结果患者癌组织中γ—GT和GSH—S—转移酶平均活性超过相应正常组织活性水平,表明机体利用硫醚氨酸途径排泄亲电子的致癌剂的解毒过程处于活跃状态,因此提示亲电子的化学致癌剂在食管癌、贲门癌等发病中起一定的作用。
【Abstract】 The Clutathione S-transferases are a major group of soluble liver proteins that are involved in the cellular detoxification of compounds. The glutathione S-transferases have been calculated to represent some 10% of soluble rat liver proteins that catalyze the conjugation of glutathione with a wide range of electrophilic compounds. The formation of mercapturic acid follows a pathway involving the reaction of a foreign compound with the sulhydryl group of glutathione catalyzed by gluta thione S-transferases to form a glutathione conjugated whose γ-glutamyl moiety is remcved by y-glu tamyl transpeptidase(γ-GT). A variety of in vitro and in vivo studies indicates that transpeptidase is, a significant physiological function of the enzyme.The content in tissue of γ-GT and glutathione S-transferases with patients of gastric and esophageal cancer were investigated and compared with those of normal tissue. The cancer tissue and normal tissue were confirmed by histopathological examinations. Though in 13 cases both y-GT and glutathione S-transferase average activity were elevated, there was no significant differences in satistical treatment. The γ-GT content in sera of these patients was within normal range. These, results support the possibility that chemical carcinogenesis play a role in the generation with malign ant tumors of digestive tract.
