化学致癌物诱导悬浮大鼠肝细胞DNA非程序合成的研究

【摘要】 <正> 细胞的非程序合成(UDS)已被广泛用于研究化学物质和物理因子的细胞遗传毒理作用,并已被认为可用作致癌物快速筛选的一种手段。一般常用人成纤维细胞、HeLa细胞和人外周血淋巴细胞,也有用人羊膜细胞为材料。然而这类细胞常缺乏完整的代谢致癌物的酶系统,因而近年来William等建立了大鼠肝原代培养细胞的UDS方法,以图保持致癌物代谢酶系的完整性。然而在肝原代培养过程中,细胞分离后必须先培养24小时,待细胞贴壁后才能用于实验;但是Michalo-

【Abstract】 This paper reported that the unscheduled DNA synthesis (UDS) in freshly suspendedrat liver cells (SRLC) can be induced by diverse classes of carcinogens, e. g. B (a) P(polycyclic aromatic carbohydrate), 2-AAF (aromatic amine), DMN (nitrosamine),nitrogen mustard and cyclophosphamide (bifunctional alkylating agents). These compoundsexerted direct and indirect carcinogenic effect leading to DNA damages that would beremoved by different repairing mechanisms. The results showed that all of these testedcarcinogens except cyclophosphamide could induce UDS in SRLC. It indicated that themetabolic response of SRLC to carcinogens is more like the situation in vivo in compare withrat S-9, and moreover the experiment can be accomplished within one day. Therefore,SRLC can be used to study cytogenetoxicology, mechanisms of carcinogenesis and as a shortterm test to detect mutagens/carcinogens.
