周期型马来丝虫感染期幼虫体外培养的继续观察

【摘要】 周期型马来丝虫感染期幼虫(L3)在含人羊膜细胞系的TC199+RPMI1640(1∶1)混合培养液中于培养后10~12d开始蜕皮,少数(9.6%)可完成蜕皮进入L4,幼虫最长可存活25d;在含人羊膜细胞系的TC199培养液及不含细胞系的TC199+RPMI1640(1∶1)混合培养液中,幼虫可分别存活19和15d,但均未出现蜕皮现象。本实验证实了培养基的组成及哺乳动物细胞系的应用对于马来丝虫体外存活、生长和发育的重要性。

【Abstract】 This paper reports the growth, devlopment and survival duration of the infective stage lervae (L3)of periodic Brugia malayi in different in vitro culture systems.A part of the infective stage larvae (9.6%) developed to the fourth stage (L4) in 1:1(by volume) mixed medium of TC199 and RPMI1640 supplemented with a feeder layer of human amnion cell line (FL). In this culture system L3 began to moult at 10th-12th days after cultivation, survied for about 25 days with the increase of 18.7% and 20.3% in length and width respectively. In TC199 medium with FL cell line and in cell-free 1:1 (by volume)mixed medium of TC199 and RPMI-1640,the larvae survived for 19 and 15 days, respectively, without moulting. The results show that the medium composition and application of the mammalian cell line are of significant importance in the in vitro survial, growth and development of B.malayi.
