分泌抗霍乱弧菌单克隆抗体杂交瘤细胞株的建立和应用

【Abstract】 Four strains of monoclonal antibody (McAb) secreting hybridoma cell were established through fusion of mouse myeloma cell and BALB/C mouse splenic cell immunized with Ogawa strain (18001) of Vibrio cholerae (biotype El Tor).These cells,no matter when they were cultured in vitro or in vivo,were shown to be able to produce McAb against Ogawa,Inaba and Hikojima strains of V.cholerae,with high specificity.The immu- noglobulin of one cell line belonged to IgG2b subclass,while those of the other three cell lines,all belonged to IgM. The cells grew well and secreted antibody consistently after 10 months of serial passage and 6 months of preservation in liquid nitrogen. The cholera SPA diagnostic reagent made of McAb (2D1) gave satisfactory result in coagglutination test of 49 strains of V.cholerae and 9 strains of other enteric bacteria.
